2.After hitting the target with two darts, you must wait for approximately three seconds for the tranquilizer to take effect. Continuously shooting and hitting the target will cause them to wake up.
Your best bet in capturing large fish before that is by upgrading your rifle to the tranquilizer tree so you have a 40% chance to tranq even large fish.
Edit: until you an actually upgrade the dart i would focus on the Tranq Rifle, more shots and still a good chance to put things to sleep.
