Unable to trigger the last quest from the village. [SPOILERS]

I just beat the game (normal ending) and I started the same file again and tried to trigger the true ending, however, I cannot make Shennong activate the last of the village quest where I need to help him defend the refugees, I saved the village the first time, I killed the yellow snake, cure the kid and max out my hp (gave him all the poisons), what else I need to do? please help

Did you give ShuanShuan all their items?
There are quite a few more even after the achievement for maxing out his relationship status pops.
If you've done an ending you should definitely have the ability to look up a guide on where all of them are and see if you missed any.

I stopped giving him more items after I unlocked the achievement "Establish a deep relationship with Shuanshuan", so you suggest finding the rest of those artefacts?

I am stuck even before that. No yellow snake for me. I have cleaned up the entire map except the abandonned cave where 1 items is missing (supposedly from the snake).

Found every last artifact, every poison, everything. None of the NPCs have any quest for me, I’ve been looking around for hours and I’m starting to think I’m locked out of the true ending :/

The yellow snake is in the forbiden cave where you start the game near the village. Get where the boar was with the flower and you'll find it.

You get the yellow snake quest by going into the Galactic Dock and speaking with the villagers. Not speaking directly to Shennong in the Pavilion.

Haven't played the game yet, but if I go through the whole game is there an auto save I can go back to in order to do quests for final boss i may have missed? how does it work if you miss him?

you get the quest after handing Ah-nong the "poison" from the village shaman after saving the village, then you go talk to the shaman, go to cave, Ah-nong again then shaman and you're done
