Nope, those slots are unlocked by Player Level, not character proficiency.

What proficiency does is still unknown outside of unlocking titles and avatars.

It doesn't, that one is unlocked for all characters by leveling up your player level.

I have no idea what the mastery does aside giving you titles for your player card.
