You can also use TransDEM to quickly build routes http://www.rolandziegler.de/ there's some videos on youtube to learn how to use TransDEM
I think you should start with what kind of landscape you want to recreate, or what kind of industry you'd like to simulate.
I've never tried it before, but if you want literal help building a route you could look into Multiplayer Surveyor. Only immediate drawback is that you must start with a base content map, instead of from scratch (at least from what I understand).
