Maybe a certain set or a lower number of them is required?
edit: ah nvm just found out that there are 13 glyphs and i thought its 12 and the last one im missing happen to be 1 of the 8 require glyphs for ending lol
Do 7 endings in NG, then 2 in NG+, and after - you'll be able to do get the last one.
I had met him first at Scorchwhere when entering from Goliathfall, but then I turned back without beating him as there were a lot of other areas I didn't explore.
Then while going through Field of Pyro, I ran into him here (not marked on Demajen's map either):

After beating him, he continued the conversation started before, which seemed odd. I thought he might have multiple battles similar to some other bosses, but afterwards he didn't show up in Scorchwhere again. So it might be possible to just beat both bosses in Field of Pyro, without visiting Scorchwhere.
Edit: I mixed up which boss is needed from Scrorchwhere. Agus can be fought only there, so the last sentence didn't make sense.
(This may not be too useful, but as it happened, in the Field of Pyro arena I found a very comfortable way to cheese Brandr: Wall double jumping in the corner with Rainfall Messenger makes you immune to most of his attacks and the droplets take care of him pretty quickly. It's interesting that the other pyro boss, Aqil, has a different, but even safer way to cheese, where he doesn't fight back.)
