Even though the Tower can become the most challenging way to play the game, it is also the best mode to get really good at the game. I would say just dive in and have lots of fun.
One tip is don't feel pressured to use what others say is the best weapon for the game, unless if you want go out of your comfort zone and challenge yourself. Just use what you are comfortable with and really enjoy. I created a save file that only has the Sidearm and the Carbine in it, and that got me in the top 116 spot out of 54,000 people at the time.
Another tip is once you get better and want to focus on getting a better high score is manage the score multiplier. You build it up as you play, but if too much time goes by before you attack the next enemy, it will begin to go back down. So move quickly and figure out tricks to keep the multiplier active. Also at end of a run, there is a terminal that gives you a breakdown of your performance and what things contributed to your final score. I would say pay careful attention to all those details and how much score each action is worth.
Some other things about Tower: It is the fasted way to unlock traits and discover new ones for your weapons, you will be spending all your time taking out tons of enemies. Plus your proficiency level goes up unusually fast in this mode. You will also discover a new kind of consumable called Disgorgers, they are basically an extremely powerful main weapon that last for 20 seconds or something like that. There are four different types, and they are only used in Tower.
Other than that I would say try to figure out what power ups you should prioritize buying first, and what artifacts you want to equip.
