The Tower of Sisyphus was just a DLC with a challenging endless mode and "House sequences" is referring to basically some interactive cut scences that explain the history of your character.
Challenge mode includes challenges that change on a daily basis that you can score highscores in and is not connected to the singlepayer.
After a year or so, they added free content in form of: The Tower of Sysyphus (an infinite tower challenge, solo only) and added multiplayer in the main story.
The coop in the main story is that you go on the world of someone, and help him progress, you get the host permanent upgrade if they have it (won't spoil you about what hey are), but it does not progress your own story, it's really an help. You get something for the help provided though (a rank, but i don't remember what for).
I finished the game when it was solo only, and honestly, the added coop mode is really a blessing, it's very fun and you can revive your mate, this can make the game a bit easier for the people finding it too hard.
IMO, the coop is worth on this game, even if this is only main story and not tower of sysyphus.
