Fortunately Picard dying of old age is not a requirement to unlock the upgrade tree, but note that you will not be able to build Defiant and Sovereign class ships until you have completed the Enterprise tree.
I just got an escort mission from the Ktarians, it was a success, and it worked (1/3).
Ive noticed this too. thats got to be a bug, doesnt make sense.

There are also two other missions I found work:
One is a science project in a minors territory, often in their capital system. Works the same way as the highway-node mission. The other is the "build up of forces" between two minors. If you side with one party, you get a "show of force" special project. There, you need 5 military ships in a neutral system. If you have four dedicated ships and the Enterprise you can trigger the mission and it will count towards the Troi-Counter.
If you don't care about cheesing, just save before requesting the mission and reload if it's not a science mission?
