Is it possible without relics? I've had no luck even with relics, but I will admit I'm not great at the game

I beat the boss of Quill's Chapter 3 without relics using a Plunder-based deck. With a little luck, you can use one to establish board control and plunder strong cards to give yourself options for when the boss drops some of his heavy hitting cards.
Here's a screenshot of the no relics win and the list of cards I used. I believe this only took one attempt with the deck, so hopefully it won't take long for you either.

@Morthgarrow Oh is the boss also 3-6? I was referring to the optional fight, the one against Dungeon Keeper who punishes you for using spells and goes for a wide board, each of which has deal 2 damage to your hero on death. I truly think it's impossible, I've given it well over 10 attempts

I take it back, I just made an all minion golem deck and lucked out to where guts bot survived the entire fight by merging into different rows a few times. What a frustrating fight.

Never assume something is impossible unless you're a master at it. It's possible but it's the hardest accolade on a hard optional sidequest boss, it is gonna require playing well outside and inside of the fight. That means identifying what makes the fight hard and countering it. For example: The fight goes for a wide board? Punish that with AOEs or by going in on big minions that little minions can't stop. The fight has some death effects that sneak a little damage in? Better bring a source of healing or survivability boost if you can't rush them down.
The easiest deck is probably golems. Summon Guts Bot and keep it alive while buffing it up with lots of cheap golems and blueprints and even the +1/+1 spells the enemy gives you. You don't need to protect every lane, once your Guts Bot gets lifesteal you can tank the enemy minion damage and death effects. You probably want Cross-Slash to counter the ghosts and other early threats. You want Augmentrons to move+buff your Guts Bot if the enemy in front of it will hurt it too much to attack through. The golem BB can bring a Guts Bot from slowly growing to huge once it's charged. Play spells smart, think about if the spell is worth it even when the enemy relic punishes you, and remember if their front row is already full then they can't get more minions and you have a good chance to spam those cheap buff spells.

Oh my apologies, I didn't realize you meant the dungeon keeper optional fight (I just assumed it was the final boss since he also has a no-relic accolade). Yeah the dungeon keeper can be a real pain, I remember getting him using a golem deck as well.
Congrats on finally clearing that one!
