If that was originally Beach under the building, it doesn't count. Placing a building converts the terrain under it to Fertile Earth.

Maybe artificial beach islands won't work. So it should just come down to spamming irrigators and scrubbers in a way that you get just the right amount of mangrove and beach where jungle can exist close enough to coral reefs.

Thanks for that. I hadn't realised that wetland counted as tier two.
So far, I understand that you have to get at least 1 contact square of the level 2 biomes - including a coastal square of the water level 2 biome - alonside the animal observatory.
I don't know if that include the squares in diagonal or not.
On my screenshot (city), this is the patch of grass 2x2, where was the animal observatory.
1. Sunflowers biome
2. Bamboo biome
3. Forest & Lagune biome regardless, as they can grow in undisputed place (buildings & water).
