Basically they all have a labeled weapon they "prefer" that allows them to level it beyond everyone else.
They also have the 2 classes also said on their tab they prefer too build up to, so just make sure you can be those and way later in game you will be able to be them.
I'm not sure how they unlock. Maybe level base? In any case you won't miss them.
In the character sheet however, if you go to affinities, each character has 2 classes there (e.g. Andrea has assassin and ranger). Upon reaching mastery level of said classes, you'll unlock the "unique" skills attached to each of them.
Class skills are unlocked with class XP - sometimes at the end of the class development, sometimes in the middle. If Class XP bar is divided in several parts then at the end of each part you'll get a class skill.
