They do not respawn as far as I'm aware, but don't fret! You don't need to have the fish to get the quest.
What you need to do is talk to the travelling merchant. You go wherever you want to meet her, and in the bottom left you see her name. Just like with any other person in the game, you can click on that name to actually talk to her.
The other things you click on are her various 'stores' or 'workshops', and there you cannot hand in this quest. It's a bit of a confusing mechanic indeed, but it serves a purpose. You can now simply sell all fish, and later still hand in the quest. She wants to know the location, so as long as you remember where you caught the fish (the game remembers it for you, don't worry) you are golden.
