Yes if max speed is your priority you'll have to mess with the gear ratios, as you can simply run out of gears otherwise.
Dunno what the theoretical top speed of the car is though.
Also if you tune it purely for max speed, the longer gear ratios might be to your detriment in other areas, like acceleration in lower gears.
There's a logical workaround for that.
As far as you didn't purchase any parts for that Lambo yourself or didn't use any other tune on it, however, if you did, grab another GTR for the next steps:
Download desired tune.
Get on any drag strip (preferably aerodomo).
Hit full throttle on reverse. Write down top speed you've reached.
Unequip tune.
Re-equip parts. Due to this being the only time you've ever purchased anything for the car, you will have an easy time rebuilding your car by equipping only parts you you own. Don't forget about rims, rim size and superchargers if you will find yourself mismatching PI.
Get back to the drag strip, hit reverse full throttle and adjust your final gear ratio in accordance with speeds reached in downloaded tune.
And viola. Parts and Final Gear Ratio is no secret anymore. Telemetry will help you get tire pressure, camber, toe and caster easily too. The rest of the settings can be discovered using Data Out Telemetry.

I'd imagine op wants to go 260mph which I would think only an engine swap would get the Diablo to that speed even with a normal gear ratio tune.
I just fully did up the lambo with the v12 race engine and got it to hit 280mph with the stock tune and I believe that's the max the 6spd can do. Can probably go faster with more gears.
