The most fun I've had so far was a 3 mage run with Sorine, Aella and Lyndal with Mobzei as pet. Pumping out hamsters and moving them around to make huge chain lightnings that hit everyone in the map and reshuffling the enemies around with Aella's typhoon was way too much fun.
I believe Rue has the best damage potential in the game, with Penetration Shot and her passive allowing her to do 4 damage to enemies in a line, twice, as long as she doesn't move (not counting dashes -- meaning you can pull off chains of Attack -> Dash -> Attack). I chose the other members based on their ability to set up such lines (Darby can be useful in the middle of the board precisely because he can't be targeted, allowing you to create choke points), or leverage the clumps of enemies that result (Sorine).
Other characters who seem to be pretty OP are Shadow (Ambush Assassin), Kalar (Spatial Manipulation Mage), and Lyndal. The position swapping stone is the best defensive item, followed by the Queen's Order wand which is great for offense. Kalar and Lyndal are appealing because they can use those abilities more than once. For pets, aside from the hamster and the ox, I also like the cat.
Rue has the most reliable high damage in the game, beaten only by the potential for the floating bunny demon's "do three damage to everything on the field" skill to trigger off under ideal circumstances.
Darby is a great character for survival and has reasonable damage potential, especially with the stealth passive skill letting him stand in the middle of the map, but I found him a bit of a struggle on harder difficulties because most of the time enemies swarm your allies and it can be hard to place him in a way that doesn't hurt anything else.
Tink is one of my personal favourites, as their main attack does proper AoE damage only to enemies when fully upgraded. That kinda power is really good at clearing the lower mobs or de-tagging shields for follow-up attacks. Plus, since their specialty is supposed to be "healer" you actually restore allied HP if any of them are in its line of fire.
As for pets, yeah, cat, hamster, and fox are the best three I've tried. Snake is actually pretty good, since the Hexed debuff (which I thought was poison for ages) actually cancels attacks, plus the snake can move again after striking. So it's a good choice for precision runs.
The floating bunny demon LOL!! Is it the pinky Lynette? I just unlocked her and started a new run. Looks cute and awesome!
I haven't unlocked many heroes or pets yet, but I do like the original squad Lyndal, Traegar and Leland, plus a kickstarter dino Pomka. The original squad gives a balanced damage and movement I think, especially for AOE damage. I have Sorine, but she's like such a dangerous lady dealing massive damage to allies as well. Snake is very handy in the trap trigger battles.
