Essentially all the enemies are some kind of horrors from the EXIT dimension which houses Diavola (True Ending final boss) who the cultist you encounter worship. Cybil leads these cultist in a ritual to bring her to our world. The exact details of what they are doing is found in Assignment Anya and this ritual is causing all sorts of problems in the mansion (for example the layout changing).
Cordelia's father Able created a portal to the EXIT dimension which lead him to being found out by the cult. (I don't known if he ever worked with them to create the portal or not). Able created a serum for Chunks (the family's pet rat) to protect everyone in case something happened to him along with a cure (which ends up only half working as Chunks only protects the mansion but will also attack the family). In the end the cultist capture him to be used as the final sacrifice. In the true ending he is saved and the cults plan thwarted for now.
All the characters motivations from coming to the mansion are explained in their cineamatics when you unlock them. Some get swept up unknowing into the mess and others are searching for the family in the mansion. There is also an arg in the games as well but it is not required to understand the plot.
