Not sure. It depends on how often you stop along the way.
It took me around 40 days the first time I reached the port. I can't recall how many hours that took.
There's an achievement for making it to the port in 21 days so you could do it in half the time I took at least.

I would add that its highly dependent on your style of playing games like these. If you want to be tactical on missions, it will increase their time and let you try to puzzle out ways to be successful as opposed to just guns a'blazin'. You also have to manage the roles on your train too, so even if you like to just blast your way through missions, if soldiers get hurt they need time to recover, and other troops to help mend their wounds. I'm on a run right now where I've still got two chapters left and I'm a ~60 hours.
