It's better to stick to one if you want to improve. If you want to just have fun and don't care about learning or improving your game, then you can play Random Civ on all maps. You will be playing at a slightly lower skill level than you're capable of, but it's a lot of fun. I have an account where I play only random and fool around for fun.
But I'll definitely be focusing mostly on my main civ for 60% of my matches

I think it is fairly balanced. It is a very asymmetrical RTS. While everyone builds the same kind of units (melee/ranged, infantry/cavalry) and have (mostly) the same mechanics, each civilization is different enough that they have different kinds of victory conditions. Some civilizations have bonuses that help them be much more aggressive than others, but I think the developers have struck a nice balance. Achieving Conqueror level 3 in ranked is possible while playing only one civilization.
I will say that every civilization feels weaker/stronger vs certain other civs, but I think the challenge of figuring out how to win from a weak position is very fun.

This website shows the winrate of every civilization in every matchup. The lowest winrate some civs have vs their counter is 42% and the highest is around 60%. So everyone is balanced -8/+10 from 50% winrate. That's actually pretty decent for having to balance 16 options.
