Yeah, you can play it 100% in VR. You can also play all of the community rooms from Steam Workshop, even though some of them may not work best in VR.
If you're asking if you could play Escape Simulator stand-alone on Quest 3 after getting it on Steam, nope. You'll have to do a separate purchase on Meta Quest store.
