I also often build a "city loop" line (which can be huge cash-cows :-) ) and these I'll start at 30/30 and bring them down in stages. I had one that got to 5/5 with large articulated busses.
I find the best practice is to monitor the usage via the stats.. show tour details section and adjust your schedules accordingly.
I work on the principle that the bus isn't making me money sitting in the depot or sitting idle between runs. :-)

By "standard bus routes" I mean a route that goes from one place to another (and possibly back again). I use this to differentiate from a "city loop" type route, where the busses just go round-and-round-and-round.

e.g. If you have an A-B route having 60 minutes round trip time from route planner, be careful on the delay that better having 10~15 minutes as buffer, or you just don't give an F about that abusing your drivers to the extreme.
Then you'll come up with 75 or 80 round trip time. If you're really short of buses (at least 2 available) or the estimated passenger really that low then 75 or 80 would be your line's headway.
Otherwise divide that 75 or 80 minutes headway with a desired amount of buses running at the same time to get the headway.
E.g. a 80-minute round trip route, suggesting peak passenger is around 300. Using Trident 2 and B9TL that capacity is around 90, thus assigning 4 buses with 20 minutes headway optimally speaking.
