Not in the old show (the one this game is based on) at least. He's not a main character, he appears just a few times.
I don't know what is the problem and why April can't just learn martial arts. I don't remember Splinter fighting that much neither and there he is in the trailer.
It would be good if they add more characters of course, but I highly doubt Casey Jones will be a playable character on the base game (wish I'm mistaken though).
It's not native to the intent of the character as-written. I hate to wade into this, but it was given to her (yes, given, you don't become a martial arts master in a year, or even five years) probably as a diversity and inclusion stunt while totally and completely forgetting April's actual purpose to the comic and the show.
Casey Jones is -far- more iconic then you depict him. He belongs here.
Irma is just too out-there even as a joke-char, Venus could be an unlock, but isn't cute like Ap, like Donny in drag.

If you want to talk about the "intent of the character as written," the original material from Eastman and Laird depicts April fighting crime as a vigilante.
Besides, Splinter had plenty of time to teach her since 1991.
This is the same as saying 'I don't see why giving superpowers to Mary Jane is a problem. Spider-Man can climb onto walls and use spider webs.'
80s TMNT is a cultural heritage, this game is based on it. The least the game can do is respect it. I'd understand if it were a different timeline.
Splinter destroys a wooden box in the intro theme of the show using badass moves. He is the ninja turtles' martial arts master. He deserves a place.

April is way more iconic as a character overall than Casey. She has basically been there since the beginning. She was the first to discover the turtles. She was always there telling you to "GO". Long before the cartoons, she was able to fight when she became the female version of "Nobody". A vigilante persona. Many iterations say that she has had training with Splinter.
The 1980's cartoon changed a LOT of stuff. While the movie meshed the two. While in those versions, she had no fighting skills. She has plenty of spirit.
Casey is important enough to exist for sure and he is not a minor character, but I would rank him below April. Yes, he can fight. He has had no training in martial arts. He is a vigilante with a street fighting style.
Splinter is the master, stays hidden while the youngin's go out and fight. Typically that is how it went in the culture. We would have to also establish if he is too old to fight, or young enough to whoop everyone's butt.
It does not surprise me that she has a more active role in the games/media. Hell, she should be trained to be a ninja master to further her reporting career. Her best friends are all fighting experts. I'm am surprised she hasn't been more combat inclined sooner.
You also have to consider how adding more characters would affect gameplay. Each turtle has their own strengths based on certain parameters like, health, speed, damage, range, and so on.
Even in The Last Ronin, her daughter is an actual resistance fighter who trained all her life in the dystopian future and Mikey swats her away like a fly.
Making April a kung fu fighter is nothing but bs.
