DLC is probably another 30-50 hours if you just want to see the story stuff, and then as many hours as you want beyond that in the endless roguelite modes or endgame challenges. The story stuff is gated behind a bit of the roguelite mode and higher difficulties so you'll spend some time preparing for those as well.
Difficulty wise, easy and normal are way easier than Nioh, hard with party members/companions is a bit easier than nioh, hard without party members (and without using some of the completely busted jobs) is maybe a bit harder than Nioh, especially mobs where enemies number much more than Nioh.
Overall I'd say to expect 30-40 hours or so baseline, and then 30-40 hours for the DLC, and then as much as you want beyond that. It's not the sort of game that is super content lite or anything if that's your main concern. If you play on easy and ignore side content you can probably beat everything in like 20 hours or so though.
