You shouldn't consider taking Mushroom until you reach the fourth gold card in chest 3 (unlocking the Druid hero).
For my first gold card in chest 2, I took Cingulata, as it represented the biggest damage increase for me.
And for my first gold card in chest 3, I actually went the poison route, as that was strong for me (while in Merchant guild) at the time. Later I regretted that, and so spent some of my crystal to undo the choice, and get some physical DPS instead. So there are times when poison is better for you than Physical DPS, but it's been the minority of the time for me so far.
