You level upboss weapons replaying the boss level. Some bosses are REALLY fun ti fight over and over again.
Others like the fool ♥♥♥♥♥♥ giant tree are boring as ♥♥♥♥ haha
There really does not happen anything. Credits roll, you get back to the main menu, you select "continue" and it is af it you had answered no. I think "yes" is just for your sake of mind.
The only point of no return is the first introduction level. If you do not pick any lore items up there, you will not be able to do it later (there was another thread about it). Other than that, everything can be replayed.
You level up boss weapons by simply playing the same level again, the boss will be alive. Though I hardly found the need for it. Have a look at the "best plague weapons" thread to decide, whether it is worth for you farming a boss mid-game.
If you cancel recall, you can go back to this memory at anytime. If you choose to fight the last boss, beating him will give you an option to pick the ending you would like.
After the ending you can choose Yes or No, but I am not sure what this does, either options sees you back at the point before you killed the last boss.
To get all the endings you will have to kill the last boss numerous times. Finishing the game also doesn't start a NG+ cycle, you can replay any boss in the game as many times as you want. So short answer is there is no point of no return.
