Does negating damage with shields and barriers still allow you to get this achievement, or do you not need to take any damage from any source?

Don't know but here's a friendly tip, use the artifact of glass (500% damage) and the artifact that makes all monsters same type.

Just get lemurians or crabs and oneshot them on first stage and voilà.
