Yes, the "Sluggys" do respawn when you reenter a cave after reloading a save game/restarting the game - dunno if they do after some time without reloading a save game/restarting the game (will check this now).

It actually makes sense and I just didn't remember the other times - when you run into that cave and come across the point were you need to blow sluggy up later on, you are not supposed to exit the cave at this point, you're supposed to find the shovel, see the building and search for a way out and maybe see Sluggy getting rid of those rocks in front of him, like I just did.
Done testing - it's triggered by running all through the cave until you stand at the abyss where you see the building on the other side (sound and a group of bats indicating you reached the point), no need to pick up anything.
