Penitent Engines are often crewed by sinners of both genders. Sisters who have truly fallen or had their faith shattered, fled from battle and so on, can also be strapped into it. However it is more commonly a Sinner who is beyond redemption from anything else.

then you don´t know the lore, these are machines for punishment. everyone can be strapped on those to get on the frontline...

they do not have to be part of the ecclesiarchy. Just anyone can receive that punishment, they are not warriors of the ecclesiarchy, they are penitents hauled towards their execution. Also hypocrisy is one of the imperiums main traits and there are more than enough male zealots and priests running around.
