No.
As long as you play on the same character, you don't lose anything.
Except quest items (but not the light)
If you re-roll your campaign using the world stone, you keep everything - You can also select difficulty when re-rolling.
Also remember, rerolling will re-generate the maps to allow spawning of alternative areas/bosses you haven't encountered before. So if you don't do that, you will be missing on a lot of content and items.
All-in-all, rerolling is what you should be doing on the long-run :)
