No.
When you buy gear or upgrade them with a character, it's permanent for that profile. If the character that purchased it dies, the next character will still have access to it.
It's the same for the castle upgrades and the runes.

Thanks, does the same thing apply to runes?

Yes. Runes and their upgrades, as well as any upgrades bought in the castle are permanent for the profile you're playing on. They can benefit all characters once you buy them and can't be lost (even after jumping in NG+)
