Could be many things, I'd first check your antivirus, a behavour shield could cause this and prevent any program from opening it. If your still having trouble download Whatsrunning (search google for that exact word) and it will tell you what process has a lock on it.

That's wierd... Every other game i have (and i have a LOT) runs smoothly...
Thanks for the advise, though... I'll try.

Well, it is more likely than not that Tripwire's software wasnt ready for the workshop update, if not try turning off your steam overlay that might help.

if you have server perks delete it its from workshop

same issues, no solution so far as far as i know, unless the perks are the issue, but i just barely bought the game, fresh install, all that good stuff. have not even looked at workshop page so... ya

If you downloaded any extra content besides Killing Floor, then deinstall it. (e.g. Defense Alliance 2 mod).

For all the times I've had trouble with L4D1,2, and other games, this suggestion has never worked. It should only be suggested for people who have deleted the game files.

The mod may have messed up some game files, verifying the game cache after deinstalling the mod replaces the corrupt files.

What you can also do is to post the KillingFloor.txt log content in here, so we can look up the file(s) that causes the steam error.

I've had this happen before but I don't know if this will help but I exited out of steam and reloaded it back up - logged in, then loaded up KF and it was fine again.

Even though the problem for me only occurs if I host with the season zed mods (any one mod at a time), here it is;

Log: STEAMSTATS: SECURITY CHECK FAILED - ../System/KF_Halloween_ZEDS.u

Here we are, the seasonal ZEDs are not whitelisted and every custom script is by default blacklisted, which disables perk levelling.
Disable the mutator or install Marco's Server perks which allow you to use perks even with blacklisted mods.
