To unlock DLC characters, you need to clear the story first or input a code at the title screen (on keyboard, that would be I J L I J L K, you should hear "Sardines!" if it works). The Kunoichi class is unlocked with Kyouko and Asuka.

Kyoko and Asuka are one of the fights available via the Dimension Guide, there is a section of it specific to the DLC characters. Kyoko and Asuka unlock Kunoichi, Prism Red unlocks the Prism Rangers, the fights against the Medic and Necromancer unlock their classes...

Once you are *actually* in Post Game, you can get Pink, Main Hero B, Deathsaber class, and Angel class after the Post Game fight against Axel.
