The main questline is the main quest of Wintersun. The questline has 3 locations, and all the sidequests can be done until you enter the last location, which is notified in as you try to enter it. Except Greybor's, who is a little impatient with his quest that has to be done before the second location.

Also, you can't enter the last location of that questline without completing your Mythic quest since you learn a thing there that lets you enter it. I won't spoil you the other locations.

Should I leave? Or there are other maps from this quest?

Wintersun? Just do it. The locations aren't back to back, you have to travel to each one separately. Completing the first location progresses the story but doesn't lock you out of anything or influence other events in other maps AFAIK.

Quests are not canceled until you agree to the Midnight Fane. There will also be a prompt to warn you that moving forward will prevent you from completing any future quests. Also there's a ridiculously long period between completing the main quests in Act III and when Galfrey arrives to unlock the Midnight Fane (like 1-2 months). So you shouldn't have any issue with getting locked out of side quests.

Also just look in your quest long to see what's under the main act quests and what is under everything else.

What? Do I have time in general? I am losing a lot of days because I didn't know where to go first...

When she comes time-related quests will fail?

I hate this kind of mechanics where developers force to play in the timming they want...

No, there's no time limit. It's a time you have to wait for her to show up, and her showing up doesn't trigger the event. You're in control of when to move on.

No, when the queen comes you can still do any leftover quests you had. There's no time limit until you yourself tell her to assault midnight fane.

Except if you are azata, after ivory sanctum and returning to drezen. You can assault midnight fane immediately without waiting for the queen. Though if you still have quests hanging you can choose to wait for the queen

So much fretting about nothing. Almost all, if not all, points of no return in the main questline will be explicitly outlined to you with the message "are you sure you want to proceed etc.".
Almost all timed missions have notifications for if they should be done before finishing chapter.
The rest requires simple logic:
One of your followers got abducted by cultists and is being carried away to sacrificial site to be killed right at this very moment.
Question to you as a player: will you spend your sweet time doing other quest, or maybe spend a week in a crusade mode? What do you think you are supposed to do?

As long as you are paying attention to the text on your screen you won't miss much of side content. You will still miss some different mythics-related or well enough hidden side missions on your first playthrough anyway, unless you follow some kind of a guide by the letter.
