Probably you already hit the knowledge cap from your laboratory.

When your workers take a break your knowledge will drop. And rise to the cap again after they return.

You will need more laboratories and library. And don't forget to upgrade them.

there is a constant drain in knowledge (your pops tend to forget stuff). so if nobody is actively gathering knowledge, you'll notice a drop. that's why I usually keep a pool of knowledge I save up just because of this.
another way to get a constant knowledge pool is to educate your pop. that gives you a base level which only drops if pops die or otherwise leave the town.
