Mastery, how does it effect gameplay?
I don't seem to notice any difference between a new skill and a "mastered" one. Anyone have an answer?

There's a little bar next to the skill. The difference is a "mastered" skill has the bar filled up and slightly orange, while a "non-mastered" skill has a white bar that fills up after it gets used in a fight. It's an aesthetic achievement that makes you feel good for filling up a bar. Maybe the developer is working on a DLC that will add in game effects.

Mastered skill gives you possibility to train your students this skill.

Mastering a skill is only really good for 1 thing, "Train with Gold" in Silver's Gym. Any skills you mastered that use Tonus, can be trained here with Gold to temporarily stop Tonus drain.

In addition it can make training students in Mick's Gym a bit easier, but it's not really that important, as the "difficulty" this is entirely decided by how many students and appropriate perks you have.
