Rome or Constantinople?
In your opinions, which is the better capital for the Byzantine Empire once all of southern italy is reclaimed?

There's a reason Constantine moved the capital. The Roman Empire was a maritime power that had no business being lead from a landlocked city.

For game play purposes, I prefer Rome. You have several very strong, very close baronies that the pope has made strong throughout the game with his endless wealth. It may not make much sense irl as thistle said, but for the game I believe Rome is the stronger capital. I much prefer to have all my personal domain to be right on top of eachother, so all the close baronies in Latium are nice.

But maritime warfare doesn't exist in CK3 so then that means Rome is better no?

Constantinople has better special buildings but later on Rome can be ridiculously developed and built up by the Pope.

I would say Constantinople is your best choice still however.

The Capital wasn't exactly moved, that was the seat of the Government temporarily a bit like Mediolanum later on; it became the Capital of the Eastern portion however but technically speaking Rome remained the Capital until 476.

Worth mentioning that while Rome isn't by the sea is extremely close to it and connected by the Tiber, which has been a very important factor in its growth and influence in the Mediterranean, Ostia Antica was the harbour and believed to be the first colonia dating back to the 7th Century BC; there was also Antium but it wasn't as important strategically speaking.

As for OP it really depends by your plans IMO, if you intend to reform Rome why not, otherwise keeping Constantinople may carry different advantages.

I've held both Rome and Constantinople. I prefer Constantinople because of the wall, and the ridiculous levies you can bring to bear.

Yeah, Rome can be heavily developed later, but by then you're probably crazy strong anyway.

The walls of Aurelian are pretty much the same as the Theodosian walls are they not?

If your religion has Constantinople as a holy site, use that. The defensive building in Constantinople is probably the best in the game the top (+5 Fort level, +100% Garrison size, +1000 garrison) Rome's defensive building gets only half that. Holding Constantinople is also a prerequisite for the "Born in the purple" trait.
You may want to keep Rome as your second duchy as the Collosseum reduces army maintenance by 5%. However, the second unique building that some provinces have always come at the cost of the the duchy building slot.

From a defensive point of view:
Reconstructing the Citadel of Aleppo or the Alhambra ruins also results in a great Fortress and constructing the Tower of London also makes for a nicely defended capital (although London can be reached by sea through the Themse).

Does it matter where your capital is in CK3? I'm not sure but I tend to move mine to the most developed/rich county I own. For roleplay purposes I'd keep the capital to Constantinople since it was historically moved due to the fact that it was closer to the main dangers the Roman empire was up against, namely the Huns and Persians and later Arabs, Turks, Mongols etc and it was also an important trading crossroad between the continents of Asia and Europe whereas Rome wasn't.

Oh so I should build castles in every available piece of land of my de jure capital's duchy then.

No, it's a completely wasted domain point. Castle (non capital of province) will have 1 less slot.

If you're stuggle with succession then it may worth to build 1-2 castles at capital as a reserve for failed succession (revoke them and use extra levies in war for heritance). But if you're know what you're doing - castles are strict no-no.
