You can right click him and find Disinherit option. This requires Renown.
You can try to force him to take the vows and become a monk, or if he is Martial educated you can try forcing him to join Holy Order.
You can murder him if you are a Sadist.
You can force him to be a Knight and then try to fight in many battles hoping that he dies in battle.
Disinherit is the easy way, but because it requires Renown, it is expensive to do when starting a game.
You don't choose who inherits, it's based on your successions laws but you may change them as the years progress.
There is a succession tab to see what heir(s) will receive what.
You may indeed disinherit in many ways described above. Death, clergy, via your dinasty...
Up to a point you'll be able to have enough resources to only pick the heir you want and make him inherit the whole thing, even with the wrong succession law.
It's always a bit risky as he may die before your ruler.
The fun part is to inbreed geniuses.
And get entire kingdoms via mariages and dynasty wars.
