You would do so, by playing as a germanic count, fabricating a claim on one of Prussias counties and going from there. Or declaring a holy war, to gain the duchy that way.
If your starting in the 867 start, there is no HRE to speak of, so you could seize Prussia and maybe expand it to become it's own kingdom by using the found a [custom] kingdom decision.
Or you could play in the later start date and play as a german ruler within the HRE and work towards getting the duchy of Prussia title.
You can declare independence from the HRE or remain a part of it/seize control of it.
You know, the usual.
As for Prussia, Prussia as you know it from EU4 does not exist, the "prussians" in which Prussia is named after exists, but they were pretty much geocide off the earth by the Teutonic Order (brutal Midieval History )
TRUE Prussians were Baltic, not German. Though about this time period is when we see German Prussia start to form after the Germans kick the Prussians out of Prussia :b

But yea, you'd have to create your own German conquest of Prussia. Culture/religion conversion is a complicated thing that's about to change when the expansion drops...
But...
Basically your council helps with that, and then you have decisions which you can exchange for prestige. I THINK it's steward that converts culture? And the priest = religion.
