So for singleplayer, you absolutely can control everything with just a joystick and 2 buttons, plus a menu button.
For two-player games, you can either use two separate controllers, or if you're using keyboard, you can set player 1 and player 2 controls to different keys on the same keyboard if you want. So player 1 can use wasd and player 2 can use the numpad, without the need to plug something else in. It's very versatile. So depending on how your custom setup works, I'm sure there's a way to get everything working nicely.
