Other than that, it is just a normal "climb back up and defeat boss" challenge with boss 66 as the target (unlike some challenge the target does not go up with each completion).
It does mean that you will probably want stronger energy and magic stats than for the basic challenge, having decent levels in NGUs helps quite a bit as well.
So you rebirth and take off the equipment orrrrr???
It takes you to a menu where you will be able to see all of the challenges, mouse over one and it will display its informations on the right.
You start a challenge by clicking on its button, it will perform a special rebirth that starts the challenge (with the exception of the laser sword challenge, that special rebirth completely resets your NUMBER to 1).
By defeating the boss that is the target of the challenge you will receive a reward.
Do note that your old NUMBER will not come back, so you will have to work your way up again depending on what adventure zone you need to re-unlock.

how can you discover every item though? Some of them need special conditions that aren't really specified
GRB = Gordon Ramsay Bolton = the game's first "titan". Titans are special bosses which appear in adventure mode. The unlock condition for the No Equipment Challenge refers specifically to the items that GRB can drop, not any of the items from any other adventure zones or titans.
For the no equipment challenge the equipment doesn't matter but you most likely want to at least have defeated UUG and unlocked beards, preferably after you put a decent amount in permanent levels for the first beard (that beard doesn't have a soft cap but around 1000 permanent levels will really help).
Pushing a bit in NGU power alpha and NGU augment will make a big difference as well and having cleared the no augment challenge should be very helpful since it greatly powers up your augments as well.
