You start the challenge like any other, it's just that during this specific challenge you receive absolutely no stat or bonus from your equipment on top of your number being reset to 1 (and your bank being wiped if you have those perks/quirks).

Other than that, it is just a normal "climb back up and defeat boss" challenge with boss 66 as the target (unlike some challenge the target does not go up with each completion).

It does mean that you will probably want stronger energy and magic stats than for the basic challenge, having decent levels in NGUs helps quite a bit as well.

So you rebirth and take off the equipment orrrrr???

In the rebirth menu, you should see a button next to "rebirth" called "challenges".
It takes you to a menu where you will be able to see all of the challenges, mouse over one and it will display its informations on the right.

You start a challenge by clicking on its button, it will perform a special rebirth that starts the challenge (with the exception of the laser sword challenge, that special rebirth completely resets your NUMBER to 1).
By defeating the boss that is the target of the challenge you will receive a reward.

Do note that your old NUMBER will not come back, so you will have to work your way up again depending on what adventure zone you need to re-unlock.

Something that might have been overlooked here - You can't do challenges at all until you've beaten Boss 58 for the first time.

Are there any guidelines for stats to be able to do this challenge in 12h or 24h?

if it's 12-24h per clear, you can probably manage that when your reach the grand corrupted tree or jake (second and third titans respectively), if it is for all 24 of them probably around walderp.

Does the challenge wipe out your entire inventory, or just negate any effects for the challenge period?

It just negates any and all effects from items for the duration of the challenge.

how can you discover every item though? Some of them need special conditions that aren't really specified

GRB = Gordon Ramsay Bolton = the game's first "titan". Titans are special bosses which appear in adventure mode. The unlock condition for the No Equipment Challenge refers specifically to the items that GRB can drop, not any of the items from any other adventure zones or titans.

So what stats should someone try this with? I want to unlock the auto-boost. The rec stats say "not sure but a lot of energy and magic power". How much is "a lot" ??

Exact stats are impossible to state since everyone will have different ones, which is why you will always see things like "at X zone/titan" or "with Y set".
For the no equipment challenge the equipment doesn't matter but you most likely want to at least have defeated UUG and unlocked beards, preferably after you put a decent amount in permanent levels for the first beard (that beard doesn't have a soft cap but around 1000 permanent levels will really help).
Pushing a bit in NGU power alpha and NGU augment will make a big difference as well and having cleared the no augment challenge should be very helpful since it greatly powers up your augments as well.

Usually you would have those requirements around Walderp.
