There is 301. Then you go to next difficulty.

Not immediately. There are other requirements besides just reaching boss 301.

Don't focus on bosses. Focus on adventure mode.

If you put too much emphasis on bosses, you slow down progress in adventure and as a consequence you also slow down overall progress. Bosses don't even matter that much. They are useful in the early game for getting exp, but farming exp means short rebirths (30 minutes) and a small number, so you're not going to higher and higher bosses. Other than early game exp, bosses are only good for unlocking whatever adventure zone you need access to.

Throughout a lot of the game you'll find that being able to push to higher bosses becomes a lot easier than being able to push into the next adventure zone or deal with the next titan. You'll often be in the situation where you've pushed enough bosses to have several adventure zones and titans unlocked that you aren't even strong enough to think about yet. That's why many will tell you that adventure mode is the main measure of progress in the game, not bosses and not the number on the rebirth screen.

But yeah... The game is structured as a climb through 3 difficulty modes, with each difficulty mode ending after climbing / re-climbing to boss 301 and beating the toughest version of the final titan that was introduced in that difficulty. A certain number of adventure zones and titans are available while in normal difficulty, and then the list expands to add a certain number of extra adventure zones and titans after that in evil difficulty, and then the list expands again with even more adventure zones and titans in sadistic difficulty.

Many of the titans let you unlock new features to use to help you climb further in the game.

And one of the features gets new layers added to it just for entering each of the higher difficulty modes.

Also note that each difficulty mode has its own separate copies of the same 11 challenges. So they have different completion counters and different unique rewards in each difficulty. The game's design intends for you to have completed most of the challenges in a difficulty mode by the time you're ready to finish it, although you are allowed to step back down to an earlier difficulty mode if you want to go back and finish the rest of the challenges later. (Many of the crazy bonuses that you build up in a higher mode will be disabled while in a lower mode, though.)
