Elves with tree-creatures become available with Forces of Nature DLC, other species and leaders are available in base-game. Forces of Nature adds 2 leaders, base-game has rest of the leaders available.
There are campaign-like game modes. In Monster Hunt there's a large set of monsters and a scenario to hunt each monster. It's not story-intense per say. There's also Stop the Army which has series of scenarios where you and your allies are attacked by a very strong approaching army and have to defend. In both of these modes, once you complete one of the challenges, the next one becomes available. Both can be played in Co-op. It's even possible to play them in PvP technically but they're not built for that as such.
Aside from these 2 that are closest to campaign mode, there's 4 more game modes, 3 of which can be played in Co-Op. These can offer a proper co-op challenge if you set the difficulty bit higher.
