Gale is probably the second most useful, he doesn't burn through all his spells like Wyll and he can learn spells from scrolls giving him the biggest variety of spells to use, and with level 3 spells now Wizard is just crazy powerful.
That said I think past act 1 we will probably get some more companions, an example is that Karlach seems very likely to become a companion down the line, though it would be strange to think you'd miss out on a Barbarian companion, should you not believe her story in Act 1 and remove that as an option later in the game.
I haven't tried sorcerer yet, there might be enough there to make them a decent option over wizard.
I think Shadowheart is a must have for how much the game just throws damage after damage at you, healing is needed, though she isn't that good at it because she is the rather weak subclass of Trickster, not sure I've ever used her class features. So cleric might be a strong option for picking as player class.
As for other companions - I'd bet money that there will be more upon release, perhaps not mindflayer suffering companions but folks who will join your party if you so desire.
Which is never.
Sure he is awesome in a fight, but as he is written he just won't ever make my team or even get to see my camp.
I fear DC's on locks and traps will increase in later acts making it important to have someone to fill out the rogue role. Again a reason I play a wizard. Sort the proficiency with background and max int and dex, but even then it hurts not to have Astarion's sneak attack.
Shadowheart is there and she ain't as obnoxious as Gale or Astarion, so her and Lae'zel are the obvious choices for me to pick. Sure Lae'zel is also a pain, but not in comparison to Gale and Astarion... At least she is upfront about her desire to kill you in your sleep.
Wyll... Well... The way my play-through's evolve he usually leaves with a sad and disappointed look on his face after some time. Perhaps I need to start killing more goblins and less druids to make him stick around. So I usually have to put up with Astarion to have a full party of four, but as soon as I am able to pick another damage dealer that doesn't run home and cry because he got a little innocent tiefling children's blood on his hands, then Astarion is gonna be handed off to a bounty hunter.
Imo hard healing in e5 is likely an option more than a 'must'. Clerics could be more profitable than just healbots.
Also (1) if you spend too much main actions on heal spell it's usually means that your party composition is not that great (2) If you spend too much actions on healing that means you even more losing efficiency of your party. It's like a loop. Healing too much is bad :D
Precisely the moment to stab her in the throat!
There are some predictions about future companions:
1) Karlach
2) Minsk, maybe as a barb
3) Datamined female charater
4) Maybe Halsin as druid
5) Maybe Jaheira?
As to current characters, running EA a lot of times, I haven't found a perfect party where most of my companions are close to desisions I'd like to make (except petting the dog).
1) Astarion is a weak skillbot. His infant behaviour is fine, but could be easily replaced with any dex character.
Also I don't like how sneak attacks implemented in this game.
2) Shadowheart is great. I don't like her build much, but she's playable.
3) Wyll is not bad, but warlocks are quite primitive class. It's not interesting for the first playthrough.
4) Alpha-gyth is annoying but sometimes fun :) As an option.
But don't like fighters as the most primitive and boring class.
5) Gale is annoying and too hungry for my precious loot.
But I want a wizard in first playthrough, to see full variety of spells. I would prefer sorcerer, but amount of spells it can learn better to leave for 2nd playthrough.
Eventually I see only shadowheart as a companion I really like :D Well, maybe Gale if he won't die from his hunger.
So hoping for interesting variations from Larian.
If your problem is sleep the WIZARD (no mages in D&D, sorry) must be elven of half elf, no "non human" in general. Also, taking from Astarion, I'm pretty sure all of them will have unique bonuses compared to custom characters, and probably you'll be able to hire mercenaires you craft yourself like in DoS2.
About rogues, Astarion is mad powerful at lv 4 with good gear. At lv 5, rogues fall behind a LOT because they don't get bonus attack, but the hand crossbows make ranged combat keep somewhat competitive and, with poisons, 1 MH and 2 OH attacks isn't really that bad.
Rogues and bards (Astarion included) have a huge benefit tough - they are the best skill monkeys. A multitude of skills (Astarion start with 7) and expertise (stealth and sleight of hand in the case of rogues). Of course, in BG3 you can always save scum, but if you don't want to do it, pickpocketing is WAY easier with those 2 classes than any other. Add to it you have an item that give +2 to those 2 skills but -1 to charisma ingame (a ring) and you have a perfect thief/assassin.
