Is a subclass something you pick on character creation or something that unlocks when you hit a certain level for your main class during play?

Depens on a class. For some classes you pick a sub-class on character creation(like Warlock or Cleric), for some it happens only after one or two level ups(like Rogue or Fighter)

Both. Depends on the class. Some subclasses unlock at level 1, some at level 2 and some at level 3.
