You can make this build DEX or STR based, both work. And you don't need to use Gloves of Dexterity or STR potions if you don't want or need them on someone else more. My go-to stat spread is:
STR build - 17 STR / 12 DEX / 13 CON (14 With Resilient) / 10 INT / 10 WIS / 14 CHA. I like taking Resilient CON, then Heavy armor master to get my STR to 18. 18 STR is good enough to hit fairly reliably when you are not taking any GWM penalties.
DEX build - 10 STR / 16 DEX / 13 CON (14 with Resilient) / 10 INT / 10 WIS / 16 CHA. This is propably the stronger of the two. Better initiative, better saves. Especially the better DEX saves combined with Aura of Warding is pretty disgusting.
For feats, Resilient CON and ASI for +2 DEX.
On both builds I use the radiating orb gear, like Luminous gloves and Callous glow ring. Ring and Cloak of Protection, Balduran's helm. On the DEX build, Belm is pretty fun end-game weapon to get a bonus action attack for three attacks per turn. With smites. And flourishes. And style points.
I know most people go by the 5 Gloom 4 Assassin 3 Champion Gloomstalker build, which 5/4 is good but at 12 ... 6 Sword bard, 3 Gloom, 3 Assassin can open with ranged slashing flourish and attack 1 target twice with each attack, that's 8 attacks hasted first round, 9 with dread ambusher 11 if you got elixir of bloodlust up ... that will burn all your inspirations in 1 round ... but 11 attacks all roughly doing something around 40-60 damage easy.
2-4 Warlock +8 Fighter, I just really love devil sight and setting up ambushes with "one with shadows" so much fun, could do something like 6 fighter 3 warlock 3 something else too or 6/2/4 etc etc, honestly I don't miss fighter 11 at all, don't need it with all my other damage flying around, I need someone to tie up enemies and fighting in darkness even allows you to go thematic when you're finishing up the fights while hasted and what not ... 1 action on One With Shadows, then double or triple attack (bonus action from GWF).
For extra actions and strategic positioning of the enemy the way I want them, I tend to have a Tempest Cleric with Hamarhraft, abusing the landing mechanics of "flight" so just by flying around the board you can reposition enemies by bouncing them with thundering shockwave into easy to aoe groups with my actions, say you got spirit guardians up, jumping around grouping em all up then drop a flamestrike or some other spell on top of the enemies ... makes a low action economy class high action economy... and I still have access to bonus action 2 turns of blade ward and bless for entire group.
Also, I like my build for Wyll, which is 6 Warlock / 6 Battlemaster Fighter. All resources come back on a short rest, so fun.
There are several multi class options for bard, warlock, paladin & sorcerer that are all insanely good as some combination of a skill monkey, tank, striker and or face of the party.
(usually grabbing smite, extra attack, meta magic and attacking with Chr are the big things to grab, though you cant really get everything so its always a choice)
Karlach as a Monk + Bear Barbarian + thief is also extremely good with tavern brawler as she will end up with 2 main hand punches & 2 off hand punches doing quite a bit of damage.
After mods ?
The bloodrager port from pathfinder is amazing and fits Karlach extremely well (its a sorcer/barbarian hybrid that gets limited spell slots and magic effects based on its bloodline while raging) (currently the undead, abyssal, devil, and fey bloodlines are in with more coming).
It also pairs well with a dip into bard so you can grab some more utility skills.
The 3.5 Duskblade conversion (there are several different mod versions) is also very good and feels like what an eldritch knight or arcane trickster always should have been (its a wizard/martial hybrid that starts off with medium armor and depending on subclass either gets additional spell slots, heavy armor or some rogue like/enhancing abilities)
The mods adding Blade dancer, Divine soul sorcerer & 2024phb sub classes like dance bard are also amazing.

Both make good assassins, the aetheric strider seems better at lethal ranged attacks (hmmm maybe combo it with blood archer). the void reaver at dual wielding.
I think I've found somebody who can take over the assassination business from Astarion.
Lae'Zel makes a rather nice 12 forceblade (mw)/8 kith'rak gish. More melee focused than the psionic warden.
I've finally tested out all 5 subclasses of the mind weaver, and I'm finding ... they all rock. Of course at first I stuck to the astral seer, which is the most "pure" psionicist, but the other subclasses are all interesting (and finally all released).
The Mystic class paralyzes you with choices of disciplines. It's too overstuffed, especially with its Expansion. But Mind Weaver seems easier to build.

Click heels + haste spore are an unholy combo to move around.
For spells you've AoE, area denial, crowd control.
With symbiotic entity you're a tank.
With sword and dagger you’ve a respectable melee damage (bypassing resistance) + silence on sussur dagger.
You start a fun guy and become the God of the Verdant Abyss, duke of decay and spore supreme.
Considering the 21 and 27 stre elixirs are in extreme abundance i hope you are not actually putting points in stre because that would be very pointless when the elixirs overwrite them.

Gloomstalker Ranger 5 + Sword bard 4 + thief rogue 3
Fighter 12.
Paladin 5, Barbarian 2, Fighter 2, Thief rogue 3.

I love how absolutely bonkers the burst damage is. Starting at level 3, it's basically a "delete one enemy of your choice" build with just Witch Bolt. And once you get up to Lightning Bolt and Chain Lightning, it's a "♥♥♥♥ that guy, and everything in his general area" kind of build.
Gear is built around maximising initiative and Spell Attack/DC:

Things become even more crazy if you fill the character up with Ilithid powers which allow them to keep killing.
Might ound strange to a lot of people, but it is how it is. :P
Only multiclass I generally use is good old gloomstalker 4/assassin 8, Otherwise all origins will mostly keep their class, apart from gale, as I usually have no need for another fullcaster in my party, so I tend to make him a monk. :P
I tend to play either college of lore bard or dragonic bloodline sorc...those are my favourite classes anyway, though I wish there was official support for Divine Soul sorcs.
After that I might play cleric or Druid, depending on the mood, sometimes even warlock, though I went away from the warlock more and more in general.
