There are several ways to start the quest. The "best" (most comprehensive) way is to go to a house near the Baldur's Gate fast-travel point. It's just south of the House of Grief. It has a Dragonborn standing outside, politely asking you not to go in.
He won't stop you from going in anyway. Do all the stuff there, and that will put you on the quest. It's a very long and involved process, but a really cool quest with a lot of loot and XP.
