Loot available down there are the Spiderstep Boots (freedom from webs), a Posion-type Robe, and the Gem/Key to open the Necromancer Book you find elsewhere in the village. It's also a method of getting down to the Underdark (there are other ways too). You can also get into the area by breaking down some walls in the blacksmith's workroom.
One other place in Act I has a pair, but interacting with them in any way isn't required at all. They're behind a gated pen below some walkways and non-hostile unless the player makes them hostile. The Ruined Temple to Selune
Act 2 has two possible instances. The first more likely, the second very miss-able.
In Act 2, you are very likely to encounter a large, intelligent, and a bit pitiable spider-like creature called a Drider. It triggers some people's arachnophobia but not everyone's.
Elsewhere in Act 2, there's a spider corpse that I could barely tell was a spider. (Gale has some funny dialog if the controlled character does something to it twice.) That spot is easily miss-able and not required.
