If you are interested, the best characters you can take story-wise are Wyll (because his father is there), Karlach (Because Gortash is there), and Gale (Because of the Netherbrain). Still the game will proceed and it is not broken if you don't take them

And regarding Mizora? Does not having Will in the party affect smth?
So when you do the shar stuff and then do the -first- fight with ketheric at the top of the tower, you'll then jump down from the top of the tower into a mind flayer colony zone(red zone) under the tower(not to be confused with the cave segment of the colony down below the dungeon... the two sections do not connect and you can only access the proper flayer colony subzone from the top of moonrise tower after ketheric's first confrontation fight).
Once down there, then you have the get mizora out of the pod option. If you dont get her out... its not good for wyll. Getting her out though will then have a progression of mizora stuff in act 3. Wyll doesnt have to be in the party but there may be cutscenes and her being in the elfsong tavern's "camp" for players that may have it requestyou put him in the party to talk with her or just auto does it(cant remember the sequence on that my first run through).
Bigger spoilers:
BUT if you DO opt to rescue the Duke, and you made other certain choices that freed Wyll from his contract and you didnt sign a new one, if you go into the challenge subzone to rescue the duke, she'll in retribution spawn a group of spiderlings right when you get the duke out of his cell that'll explode and potentially kill the duke if you dont prepare properly(its a hectic rescue challenge because when you enter the subzone you have a very very limited number of turns to get him out of his cell and back to the submarine or you fail. So if you end up having the spiderlings spawn, best know where they'll spawn (right outside his cell) and place lots of health potions on the ground around him. The explosions will auto heal him in splashes and keep him from dying if placed properly.
Also the number of turns you have for the rescue depends on the difficulty setting you choose too with a higher dif meaning less. So movement boosts, arrows of displacement, gate/misty step and other movement things are vital to helping get the duke out.... BUT also you may be wanting to get the gnomes imprisoned there out too which is a side quest part for good player runs and helping make the foundry later in act 3 go easier(dont attack the foundry first in other words).
Yes, I already saved Mizora, that's the point - I discovered it was her (not some devil she said it would be) and was like "shesh, I should have taken Will along". OK, so they will have a converstion later anyway. Good to know, thanks!
Yes, if you got wyll out of his contract, she'll try to make him reup his contract to tell you where the duke is being held though you can find out other wise. That's part of what I added just now to my other post in regards to what happens if you dont reup his contract.
Also if you dont reup it and karlach is still part of your camp and you dont opt to make her a sacrifice in the end game choices, she and wyll will have specific differences in end game scenes.
