In his particular case, you kinda do have to be a warlock. His whole character arc is about making a deal with the devil so he can arise as a hero and protect those in need. Well, a bit more complicated, but that's the seed of it. And it was all because he wasn't skilled in enough otherwise. So yeah, Wyll - I think - kinda has to be a Warlock for his story to work.
Yes you can change his class, though it's not 100% clear how it will affect their origin story as it unfolds, several content creators seem to think that it will lock you out of some dialogues and situations if you change their intended class.
