Do proficiencies stack?
Is there a way to stack either the same proficiency, or different ones that effect the same item?
For example 2h sword and longsword (used as 2h weapon).

Proficiency is a modifier you add to things you are proficient in. At level 1 its a +2 and icreases with levels. You cant stack multiple proficiency bonuses as it is added to things you are good in and not gained through being good at stuff.

No, a longsword being used in two hands is still a longsword, proficiency bonuses don't stack. It's a static thing, you're either proficient or you're not.

Well, kind of. There's a unique ability that some classes can get (rogues and bards, mostly) called Expertise, that allow you to double your proficiency modifier on a select number of skills of your choice. Currently however, Expertise doesn't seem to be in the game.

My guess is they might add in Expertise whenever they add in bard, whether that's in an update or at release.

There is no "two-handed sword proficiency". There's proficiency in greatswords. Wielding a longsword in two hands doesn't make it a greatsword.

Proficiency bonus no longer comes from the specific skill you are proficient in, instead you get a flat 'proficiency bonus' based on your level (starting at +2 at level 1) and this increases over time as you level up (to a maximum of +6). Your proficiency bonus is added to any skill roll you are proficient in, as well as any attack roll with a weapon you are proficient in, and is sometimes used in some niche cases with class based abilities.

If you are a fighter you are proficient in all martial weapons. All that means is your proficiency bonus is added to attack rolls when using that weapon. Proficiency starts at +2 and goes up +1 every 4 levels to a maximum of +6.

So, if you are using a greatsword or a longsword, you roll a 20 sided die, add your strength modifier and, if you are proficient, your proficiency bonus.

If you have a strength of 16 the modifier is +3. So a level 2 fighter would swing the sword, roll a 20 sided die, add +3 for strength and +2 for proficiency. If the total meets or exceeds the enemies AC (armor class) you hit and then you roll damage, the die determined by the type of weapon plus appropriate attribute modifier.

Strength handles all melee weapons.

Dexterity handles all ranged weapons.

Melee weapons with the finesse feature can use either strength or dexterity depending on whichever one is higher (daggers, short swords, rapiers).

Casters doing ranged attacks use proficiency and whatever attribute modifier is important for their class. Clerics and Druids use wisdom, Sorcerers and Warlocks use Charisma and Wizards use Intelligence.
