Trains will not pass signals if there is a train ahead in the next block.
Signals are two-way by default, you can set individual signals to be one way, if you do not want traffic to run the opposite direction on a specific track.
While tracks may have multiple signals, usually only one or two set to one way are required to achieve the desired result, in any given section of track.
Signals in this game aren't "one-way" or "two-way".
A "two-way" signal would be a pole with signals in both directions. Some games have those, this game does not.
They are ALWAYS one way only. The difference is that per default trains can and will use the line in the "other" direction if no other path can be found.
So in this game the choice is between signals that CAN be passed in the opposite direction (the direction the signal is not facing)... and signals that forbid such use.
In the first case, the game tries its best to respect the direction of signals, but it will run trains against your signals if it can't find any other path to the destination. It will not run trains past these signals if there's a path that doesn't pass signals "the wrong way".
In the second case, the game will post an error message "path not found" if the only path would mean having to pass a one-way signal.
Is teribly explained and the arrow should be going both directions.
Create a double-tracked line on a brand new map.
Signal one track one way and the other track the other way. (Using the default signals that doesn't block "wrong way travel")
Make sure all signals on a track point the same way! (I know several countries prototypically signal both tracks on double-tracked lines in both directions. This is realistic and looks nice, but doesn't work in this game)
Trains will respect your preferred direction. No matter which way the line curves, disproving your theory.
If trains don't respect your signals it is because there is no other path. Maybe you made a mistake building the line somewhere? Missing pieces of rail? Disconnected switches? Isolated instances of signals that force one-way travel?
Maybe you accidentally built signals in both direction on that stretch of rail? (the only way you can tell the game to use one track in one direction and the other track in the opposite direction is to unfailingly build all signals on a given track in only one direction)
a) there are only signals for trains, so the only "vehicles" you could be talking about are trains
b) trains WILL disrespect a "two way" signal, but only if that's the only path the game can find. (By "two way" I mean a signal that allows "wrong way" travel, not a true two way signal with lights in both directions)
I don't use them much but I believe you can also set buoy direction for ships though I haven't tested for any vehicular 'disrespect'.😉
All my stations have a crossover point so trains can switch lanes, they just do whatever.

You can also upload the save to the Worksshop, done via the "Share" button in the Load Game menu.
Then make them one-way.
Automatic track assignments work reasonably well in simple setups, but the more complex the track setup is, the higher the chances are the game will automatically create the line in a way you do not want.
One way signals help, but are not mandatory.
You can always reroute lines manually via waypoints or signals, either as you make the line, or via the Line Manager at anytime.
In A>B>C+ lines with multiple stations in between termini, you can manually set platform assignments, preferrably after the basic layout of the line is complete.
One way signals are definitely not needed.
