Never noticed long waiting time, but I always set the timer to indefinite, since I'm not sure if the timer counts ingame time or real time (and 1 minute real time would be very long...).
depending how long your line is and how many vehicles are there, the spacing mechanism can last up to 2 times the drive durration.

Setting the waiting time to anything at this point means nothing, has no effect..
adding more carriages at this time can in some circumstances work but not generally, usually its time to upgrade when this happens to a 5 carriage.
The game likes to auto-space vehicles up to a certain point, and has nothing to do with the wait time, which only applies if a vehicle is sitting waiting at a platform partially empty. Once frequency of a line gets good enough, vehicles will load and go, but loading/unloading of early game trucks is going to be the bottleneck, and the only way to increase the throughput is to setup an A1-B1-A2-B2 sort of back and forth line where the line alternates between multiple platforms, but you have to be prepared for horse carts getting in each others way, and if you're particularly unlucky, civilian traffic clogs the roads causing even more problems.
1. Make sure you have multiple platforms and if you are using more than 4 - 5 trucks distribute it to different platforms so that it doesn't clog.
2. Don't set the timer at 1 min interval. In fact, don't set the timer at all. I was surprised to find that when I removed the timer, the trucks seem to operate more quickly.
Also, I feel we need more functionality it terms of determining the route and more options to control how trucks enter the cargo platform and how they exit.
